How It Works

The Target

NNMT (Nicotinamide N-methyltransferase) — an enzyme that diverts nicotinamide away from NAD+ production. NNMT is overexpressed in fat tissue of obese individuals.

The Mechanism

By blocking NNMT, more nicotinamide is available to produce NAD+ — a critical coenzyme for cellular energy metabolism. Higher NAD+ levels activate sirtuins and boost mitochondrial function in fat cells, shifting them from energy storage toward energy expenditure.

Why It Matters

Preclinical studies showed NNMT inhibition reversed diet-induced obesity in mice without changing food intake, suggesting the metabolic shift alone can drive changes in body composition.
